Abstract:
In one aspect, the invention relates to a method for fixing a short nucleic acid in a biological sample. In another aspect, the invention relates to a method for detecting a target short nucleic acid in a biological sample. The method includes contacting the biological sample with an aldehyde-containing fixative, and subsequently contacting the sample with a water-soluble carbodiimide. In a further aspect, the invention relates to a kit for fixing a short nucleic acid in a biological sample. The kit includes a support substrate for holding the sample; an aldehyde-containing fixative; and a water-soluble carbodiimide.

Abstract:
The present invention provides novel methods for exponential amplification of labeled nucleic acids with high sensitivity and specificity. In one aspect, the invention includes a method for exponentially amplifying the signal of a fluorescently labeled primary click-amplifying FISH (clampFISH) probe. In another aspect, the invention includes a method for labeling a target nucleic acid in a sample. In yet another aspect, the invention includes a method for detecting a fluorescently labeled target nucleic acid in a sample. The present invention also provides a kit for use with the methods of the invention.

Abstract:
Methods for grading carcinomas are provided as well as kits therefor. A method is provided for grading a carcinoma, or suspected carcinoma, comprising obtaining a sample of the carcinoma or suspected carcinoma and contacting the sample with one or more reagents so as to identify LacNAc or Gal.beta.1,4GlcNAc expression on cell surfaces thereof, quantifying the identified LacNAc or Gal.beta.1,4GlcNAc expression, and comparing quantified LacNAc or Gal.beta.1,4GlcNAc expression to one or more predetermined control values, and assigning a grade to the suspected carcinoma based on the quantified LacNAc or Gal.beta.1,4GlcNAc expression being in excess of, or less than, the one or more predetermined control values. Also provided is a method for identifying a sample from a subject as a carcinoma sample. Also provided is a kit for grading carcinoma samples comprising a LacNAc-specific or Gal-1,4GlcNAc-specific glycosylation enzyme and a synthetically-labeled sugar molecule and written instructions for use thereof.
