Abstract:
Provided herein are SIRT4 compositions and methods of use thereof. The invention provides functional information for use in the identification and design of compounds that modulate SIRT4 enzyme activity (e.g., inhibition of fatty acid oxidation, ADP ribosylation, and/or downregulation of glutamate dehydrogenase), and to the compounds identified by such methods and the research, diagnostic and therapeutic uses of such compounds.

US Patent:
20070099830, May 3, 2007
Filed:
Apr 21, 2006
Appl. No.:
11/409170
Inventors:
Leonard Guarente - Newton MA, US
Marcia Haigis - Stoneham MA, US
International Classification:
A61K 48/00
A61K 38/17
C12Q 1/48
US Classification:
514012000, 514044000, 435015000
Abstract:
It has been discovered that Sirt4 possesses an ADP-ribosyltransferase activity. Sirt4 is localized to mitochondria, where it binds to and regulates the activity of proteins such as glutamate dehydrogenase. The ADP-ribosyltransferase activity of Sirt4 is important for the regulation of biological functions such as insulin secretion. Methods of screening for compounds that modulate the expression or activity of Sirt4 are provided. Also provided are methods of modulating insulin secretion, treating metabolic disorders, and treating neurodegenerative disorders by modulating the expression or activity of Sirt4.
