Methods Of Analyzing Polymers Using Ordered Label Strategies

View page
US Patent:
6403311, Jun 11, 2002
Filed:
Aug 13, 1999
Appl. No.:
09/374415
Inventors:
Eugene Y. Chan - Boston MA
Assignee:
US Genomics - Woburn MA
International Classification:
C12Q 168
US Classification:
435 6, 435375, 435 5, 2041828, 935 77, 935 78, 436 94, 548416, 549382, 546 33
Abstract:
The invention relates to methods and products for analyzing polymers. The polymers are analyzed by reconstructing sequence information from population data sets. The data sets include information about polymer dependent impulses arising from the polymers. The invention is also a method for linearly analyzing polymers by assessing the intensity of a signal arising from the polymer. The signal is generated as units and/or units specific markers pass a fixed station. The quantitative intensity of the signal is proportional to the number of units and/or unit specific markers giving rise to the signal.

Methods And Apparatuses For Characterization Of Single Polymers

View page
US Patent:
6762059, Jul 13, 2004
Filed:
Feb 14, 2001
Appl. No.:
09/783472
Inventors:
Eugene Y. Chan - Brookline MA
Lance C. Gleich - Somerville MA
Parris S. Wellman - Hillsborough NJ
Assignee:
U.S. Genomics, Inc. - Woburn MA
International Classification:
G01N 2101
US Classification:
436164, 436 86, 436 94, 436172, 435 6
Abstract:
The present invention relates to methods and apparatuses for characterization of single polymers. In particular, the invention relates to methods and apparatuses for determination of the velocities of single elongated polymers. Center-of-mass velocity, center-to-center velocity, end-to-end velocity and rise-time velocity are determined using time-correlated measurements of single elongated polymers in two or more detection zones. The invention also relates to methods of determinating lengths and molecular masses of single polymers and to methods of determining the distance between landmarks on a single polymers based on their velocities. The invention further relates to methods of single-molecule DNA restriction fragment analysis.

Methods Of Analyzing Polymers Using A Spatial Network Of Fluorophores And Fluorescence Resonance Energy Transfer

View page
US Patent:
6772070, Aug 3, 2004
Filed:
Feb 15, 2001
Appl. No.:
09/783930
Inventors:
Rudolf Gilmanshin - Waltham MA
Eugene Y Chan - Boston MA
Assignee:
U.S. Genomics, Inc. - Woburn MA
International Classification:
G06F 1900
US Classification:
702 19, 435 6
Abstract:
The present invention relates to methods and apparatuses for analyzing molecules, particularly polymers, and molecular complexes with extended or rod-like conformations. In particular, the methods and apparatuses are used to identify repetitive information in molecules or molecular ensembles, which is interpreted using an autocorrelation function in order to determine structural information about the molecules. The methods and apparatuses of the invention are used for, inter alia, determining the sequence of a nucleic acid, determining the degree of identity of two polymers, determining the spatial separation of specific sites within a polymer, determining the length of a polymer, and determining the velocity with which a molecule penetrates a biological membrane.

Optically Characterizing Polymers

View page
US Patent:
6790671, Sep 14, 2004
Filed:
Aug 13, 1999
Appl. No.:
09/373822
Inventors:
Robert H. Austin - Princeton NJ
Jonas O. Tegenfeldt - Princeton NJ
Eugene Y. Chan - Boston MA
Assignee:
Princeton University - Princeton NJ
International Classification:
C12Q 168
US Classification:
436172, 436 94, 436164, 436177, 422 8201, 422 8205, 422 8208, 422 8212, 204155, 204603, 2504581, 2504611, 2504612, 356344, 435 6, 435808
Abstract:
The invention relates to new systems, methods and products for analyzing polymers and in particular new systems, methods and products useful for obtaining sequence information from polymers. The invention has numerous advantages over prior art systems and methods used to obtain sequence-related information. Using the methods of the invention the entire human genome could be analyzed several orders of magnitude faster than could be accomplished using conventional technology. In addition to obtaining sequencing information for the entire genome, the systems, methods and products of the invention can be used to create comprehensive and multiple expression maps for developmental and disease processes. The ability to analyze an individuals genome and to generate multiple expression maps will greatly enhance the ability to determine the genetic basis of any phenotypic trait or disease process.
