Methods And Pharmaceutical Compositions For Inhibiting Tumor Cell Growth

View page
US Patent:
6552055, Apr 22, 2003
Filed:
Sep 4, 1997
Appl. No.:
08/923346
Inventors:
Bruce M. Spiegelman - Waban MA
Elisabetta Mueller - Boston MA
Pasha Sarraf - Boston MA
Soner Altiok - Boston MA
Peter Tontonoz - San Diego CA
Samuel Singer - Brookline MA
Assignee:
Dana-Farber Cancer Institute - Boston MA
International Classification:
A61K 3144
US Classification:
514369
Abstract:
The present invention is based on the finding that activation of PPAR plays a key role in inducing growth arrest and differentiation of certain actively proliferating cells. We show that administration of PPAR agonists, such as thiazolidinedione ligands (TZDs), is effective both in vitro and in vivo at inhibiting the proiferation of such cells.

Filed:
Sep 17, 1999
Appl. No.:
9/319769
Inventors:
Bruce M. Spiegelman - Waban MA
Soner Altiok - Cambridge MA
Elisabetta Mueller - Boston MA
Pasha Sarraf - Boston MA
Peter Tontonoz - San Diego CA
Assignee:
Dana-Farber Cancer Institute - Boston MA
International Classification:
C12Q 134
C12Q 100
G01N 3353
US Classification:
435 71
Abstract:
A method for inhibiting proliferation of a PPAR. gamma. -responsive hyperproliferative cell which comprises the step of contacting the cell with (I) an inhibitory amount of a PPAR. gamma. agonist and (II) a MAP kinase inhibitor is disclosed. A method for treating or prophylactically preventing in an animal subject a disorder characterized by unwanted proliferation of PPAR. gamma. -responsive hyperproliferative cells which comprises administering to the subject (I) an inhibitory amount of a PPAR. gamma. agonist and (II) a MAP kinase inhibitor is also disclosed. Pharmaceutical compositions comprising a therapeutically effective amount of a PPAR. gamma. agonist and a MAP kinase inhibitor are disclosed for use in the methods.
