Abstract:
An electroporation buffer comprising: a solvent; a sugar; a chloride salt; and a buffering agent. In certain embodiments: the solvent is water; the sugar is glucose or mannitol; the chloride salt is potassium chloride (KCl) or magnesium chloride (MgCl); and the buffering agent is sodium phosphate, 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) and/or dimethyl sulfoxide (DMSO). A method of electroporation, the method comprising applying an electric current to a suspension comprising: isolated eukaryotic cells; a biological material that is exogenous to the cells; and the aforementioned buffer. A recombinant cell produced using such a method. An electroporation apparatus comprising: one or more chambers; one or more pairs of electrodes configured to generate electric fields within the one or more chambers, wherein each electric field corresponds to one chamber; and a flow channel. A method for electroporation comprising utilizing the aforementioned electroporation apparatus.

Abstract:
Electroporation is a technique in which an electrical field is applied to cells in order to increase the permeability of the cell membrane. This allows for chemicals, drugs, and/or macromolecules such as proteins and nucleic acids to be introduced into the cells. Embodiments relate to an electroporation apparatus. The electroporation apparatus comprises: a plurality of chambers configured to store a plurality of cells during an electroporation process; a plurality of electrodes configured to generate a plurality of electric fields within the plurality of chambers during the electroporation process, each electric field of the plurality of electric fields corresponding to one chamber of the plurality of chambers; a flow channel configured to transport the plurality cells during a cell collection process after the electroporation process; and a plurality of valves connecting the plurality of chambers to the flow channel.

A keepout zone detection system and method for a hazardous area, such as the workspace around a power tool, that includes a safety device that controls a safety event, such as disabling the power tool, and a keepout zone detection device that includes at least one imager for imaging a workspace associated with the power tool. A processor dynamically determines the keepout zone specific to the power tool based on information received from the imager(s) or by referring to a predetermined keepout zone stored in a memory, where the processor includes a pre-trained neural network that processes the received images to further identify the presence of human appendage(s) within the keepout zone; and a communication device that communicates with the safety device to control the safety event, such as disabling the power tool, based on the determination of the presence of the human appendage within the keepout zone.

A dynamic rollover zone detection system that is easily attached to mobile machinery and dynamically detects a rollover zone around the mobile machinery. The system detects humans and objects within the rollover zone using one or more pre-trained machine learning models in order to prevent accidents and potential collisions between machinery and people or objects in the rollover zone. If a human or obstacle is identified in the rollover zone, an alert or warning is provided and/or the mobile machinery is disabled.

An apparatus for stabilizing a prosthetic heart valve within an atrioventricular valve includes a collapsible and expandable stent having an inflow end, an outflow end, and struts forming a plurality of cells connected to one another in a plurality of annular rows around the stent. The stent further includes a collapsible and expandable valve assembly secured to the stent. The valve assembly includes a cuff and a plurality of leaflets having an open configuration and a closed configuration. A cord connected to the stent includes an attachment device adapted to couple the stent to heart tissue of a patient. A method of stabilizing a prosthetic heart valve within an atrioventricular valve annulus is also described.
